Unfortunately, there's not enough information in post to for me tomake a determination. Jack the rear of the truck up and put it on stands and put blocks in front of the front tires. Have an assistant run the trans thru the gears and isolate the noise that way.

Something else to consider is that it's just tire noise. My truck starts whining when the tires get about 8K on them since last rotation.

Must be something thats spinning under truck. Only hear whine when accelerating. No whine whlie coasting and applying any gas. Maybe u joints? Doesn't seem to transmission since it happens gears 3to5. Wheel bearings are cool off highway.

Unlikely that it's u-joints, they vibrate when the start going out and I've been told by a driveline shop that Nissans use really good product right from the factory.
Your first plan seems the most likely. You may want to open the hood and make sure it's not the timing chain doing the whining as well if you can't come up with an under vehicle root cause.
